Description
ITH-12575 is an inhibitor of the mitochondrial Na⁺/Ca²⁺ exchanger (mNCX) that modulates mitochondrial calcium homeostasis, and at low micromolar concentrations ITH-12575 reduces Ca²⁺ influx through CALHM1 channels, supporting its use in studies of mitochondrial signaling, neuronal excitability, calcium-dependent metabolism, and neurodegenerative disease mechanisms.
In vitro
In models utilizing SH-SY5Y neuroblastoma cells and bovine chromaffin cells, ITH-12575 effectively mitigated cytotoxicity induced by Veratridine and high concentrations of potassium (K+). The compound prevented the rapid efflux of Ca2+ from mitochondria during depolarization, thereby dampening toxic cytosolic Ca2+ spikes and preventing cell death [1].

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
