Description K-80003(TX-803) is a potent retinol-like X-receptor-alpha regulator that inhibits TRxrα-dependent Akt activation and tumor cell growth.
In vitro In MCF-7 cells, cotreatment with K-80003 inhibits the TNFα-induced colocalization of tRXRα with p85α in the cytoplasm, leading to tRXRα nuclear localization. Western blotting reveals that the K-80003-stabilized tetrameric form of tRXRα is exclusively present in the nuclear fraction, while the tRXRα monomer is distributed in both nuclear and cytoplasmic fractions [1].
In vivo K-80003, a potent inhibitor of AKT activation by all-trans-retinoic acid, exhibits enhanced efficacy in inhibiting tRXRα-dependent AKT activation and tRXRα tumor growth in animals. K-80003 has high affinity for RXRα but lacks COX inhibitory activity [2].
