Description ALS-8112 is a inhibitor of respiratory syncytial virus (RSV) polymerase. The 5'-triphosphate form of ALS-8112 inhibits RSV polymerase (IC50: 0.02 μM).

In vitro ALS-8112 is a comprehensive inhibitor of RSV (respiratory syncytial virus) replication across various strains when assessed in vitro. Upon entering epithelial cells within the respiratory tract, ALS-8112 undergoes phosphorylation to produce an active nucleoside triphosphate form (ALS-8112-TP) that has a half-life of approximately 29 hours. This metabolite executes its antiviral action by terminating the extension of the viral RNA chain, effectively inhibiting RSV replication. Specifically, ALS-8112-TP targets and interferes with the RNA transcription activity of the RSV–RNP complex in a dose-dependent manner, displaying an inhibition concentration half-maximum (IC50) of 0.020 ± 0.008 μM. Thus, the 5'-triphosphate version of ALS-8112 distinctly halts the function of the RSV polymerase, leading to the cessation of RNA synthesis through chain termination, marking its specificity and efficacy as an antiviral agent against RSV.
