| In vitro | Agomelatine and S 21517 have relatively strong affinity for 5-HT2C receptors, with Ki values of 0.21 μM and 0.13 μM, respectively, while their metabolite 3-Hydroxy agomelatine has a lower affinity, with a Ki value of 1.8 μM, which is about 10-fold lower than that of the former two. All three are antagonists at the 5-HT2C receptor, and the order of potency is S 21517 > agomelatine > 3-Hydroxy agomelatine. [1] |
