In vitro | Topo I-IN-1 (Compound 14d), at concentrations ranging from 0-50 μM over 48 hours, demonstrates significant antiproliferative effects on human cancer cells, including A549, HCT116, MCF7, and SK-MEL-28, with lower toxicity toward normal rat kidney epithelial cells (NRK). Specifically, in vitro cytotoxicity assays reveal IC50 values of 2.33 ± 1.52 μM (A549), 2.85 ± 0.83 μM (HCT116), 26.23 ± 2.48 μM (MCF7), 3.86 ± 0.33 μM (SK-MEL-28), and 29.99 ± 0.95 μM (NRK), indicating a potent and selective toxicity profile against cancer cells over normal cells. Additionally, at lower doses (0-4 μM) for the same duration, Topo I-IN-1 exhibits a dose-dependent increase in apoptotic activity in A549 cells, characterized by typical morphological changes, alongside inducing reactive oxygen species (ROS) generation and inhibiting cell migration, further elucidating its mechanism of action against cancer cells [1]. |
