| In vitro | Topo I-IN-1 (Compound 14d) exhibits significant antiproliferative effects on human cancer cells, including A549, HCT116, MCF7, and SK-MEL-28, while showing lower toxicity to normal rat kidney epithelial cells (NRK). In vitro cytotoxicity assays determined IC50 values of 2.33 ± 1.52 μM (A549), 2.85 ± 0.83 μM (HCT116), 26.23 ± 2.48 μM (MCF7), 3.86 ± 0.33 μM (SK-MEL-28), and 29.99 ± 0.95 μM (NRK), indicating a potent and selective toxicity profile. At lower doses (0-4 μM), Topo I-IN-1 induces dose-dependent apoptotic activity in A549 cells, marked by morphological changes, ROS generation, and inhibition of cell migration, further elucidating its anticancer mechanism [1]. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
