Description
SynuClean-D (SC-D) is an α-synuclein aggregation inhibitor that prevents fibril propagation, disrupts mature amyloid fibrils, and abolishes dopaminergic neuron degeneration in an animal model of Parkinson's disease.
In vitro
SynuClean-D prevents fibril propagation in protein-misfolding cyclic amplification assays and decreases the number of α-synuclein inclusions in human neuroglioma cells. SynuClean-D significantly reduces the aggregation of wild-type α-synuclein and the familiar A30P and H50Q variants in a substoichiometric molar ratio. SynuClean-D can bind to cavities in mature α-synuclein fibrils[1].
In vivo
SynuClean-D-treated worms show decreased α-synuclein aggregation in muscle and a concomitant motility recovery. SynuClean-D is able to rescue dopaminergic neurons from α-synuclein-induced degeneration[1]. In two parkinson’s disease (PD) Caenorhabditis elegans models, SynuClean-D expresses α-synuclein either in muscle or in dopaminergic neurons and significantly reduces the toxicity exerted by α-synuclein[1].

Stock Solution Preparation:

Dissolve 2 mg of the compound in 100 μL DMSOTargetMol | reagent to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
