In vitro | SRI-41315 demonstrates cytotoxicity with CC50 values exceeding 50 μM in FRT and 16BE14o- cells, highlighting its enhanced potency and efficacy, particularly noticeable in FRT cells and correspondingly in 16HBE14o- cells. At a concentration of 5 μM over a 20-hour exposure, SRI-41315 effectively depletes eRF1 levels via a proteasome-mediated degradation pathway, as confirmed by Western Blot Analysis in CFTR-G542X 16HBEge G542X cells. Interestingly, the eRF1 degradation induced by SRI-41315 is inhibited by (S)-MG132, a proteasome inhibitor, but remains unaffected by the neddylation inhibitor MLN4924, suggesting a specific proteasomal degradation mechanism. |
