Description
Progerinin (SLC-D011) is an orally active progerin-lamin A binding inhibitor that alleviates nuclear deformation and growth arrest, suitable for studying Hutchinson-Gilford progeria syndrome (HGPS).
In vitro
In HGPS patient-derived fibroblasts (e.g., AG11498), Progerinin (100 nM, 3 days) significantly reduced progerin expression and induced the expression of H3K9me3 and cell cycle protein Cyclin B1 [1].
In various HGPS cells (e.g., AG11513, AG11498), Progerinin (2 μM, 7 days) decreased progerin and p16/INK4A expression while increasing H3K9me3 levels [1].
In vivo
In LmnaG609G/G609G mice, intraperitoneal injection of Progerinin (20 mg/kg, twice weekly) increased body weight and extended lifespan to 21 weeks; oral administration (50 mg/kg, once daily) also increased body weight and prolonged lifespan [1].

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
