| In vitro | Treatment with OSMI-3 (Compound 2b) at concentrations ranging from 20 to 50 μM for durations of 4 to 24 hours in HCT116 cells significantly decreases O-GlcNAc levels, demonstrating more sustained cellular effects compared to OSMI-2[1].In cells treated with OSMI-3 (Compound 2b), there is an observable decrease in HCF-1 cleavage products and an increase in uncleaved HCF-1. Given that OGT knockdown is known to reduce cell proliferation, the impact of OSMI-3 on cell growth over 96 hours is monitored. Although there is no evidence of apoptosis, a reduction in cell growth over time is evident, consistent with the results observed with OGT knockdown[1]. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
