Description
OncoFAP acetate is the highest-affinity small organic fibroblast activation protein (FAP) ligand reported to date, exhibiting a dissociation constant of 680 pM as measured by fluorescence polarization. OncoFAP derivatives, when administered intravenously in both fluorescent and radiolabeled forms, selectively accumulate in FAP-positive tumors while sparing normal tissues, enabling OncoFAP acetate to serve as modular components for targeted therapeutic delivery of radionuclides, CAR-T cells, or cytotoxic agents in oncology studies.
In vitro
OncoFAP acetate exhibits low cytotoxicity toward NHC cells (IC50>2.5 mM) [1]. OncoFAP acetate (57-285 μM, 1-4 h) significantly enhances the cellular uptake efficiency of NHC cells [1]. Additionally, OncoFAP acetate demonstrates the ability to improve corneal permeability [1].
In vivo
OncoFAP acetate (570 μM, administered in three doses at 10-minute intervals) was delivered via the conjunctival route, which ensured prolonged retention of OncoFAP acetate in ocular tissues [1]. In the LS174T tumor-bearing mouse model experiment, intravenous injection of OncoFAP acetate at a concentration of 12.5 μM significantly increased the retention rate of single-chain antibodies in the model [2].
