Description
Neoseptin-3 is a novel small molecule agonist of mouse mTLR4/MD2 with EC50=18.5 μM that does not activate human TLR4/MD2.

In vitro
Neoseptin-3 is a synthetic small-molecule TLR4-MD-2 agonist that induces strong TNF-α expression in RAW264.7 cells (EC₅₀ = 5.9 ± 0.4 nM), comparable to LPS. Neoseptin-3 also activates TLR4 reporter systems and promotes NF-κB activation[1].
In vivo
In mice, intraperitoneal injection of Neoseptin-3 (2.5 mg/kg) significantly increases serum TNF-α levels, demonstrating in vivo TLR4 agonist activity[1].

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
