Description
NAADP receptor modulator-1 is a cell-permeable modulator of NAADP receptors. It competes with NAADP for binding sites and inhibits NAADP-induced Ca2+ release.
In vitro
NAADP receptor modulator-1 (Compound 3) effectively and selectively inhibits NAADP-induced Ca²⁺ release in Lytechinus pictus egg homogenates with an IC₅₀ of 15 μM, showing partial agonist activity at concentrations exceeding 100 μM. It exhibits weak competitive binding to [³²P]NAADP in Lytechinus pictus egg homogenates, having an IC₅₀ of 90 μM. NAADP receptor modulator-1 (10 mM) permeates the cell membranes of intact Lytechinus pictus eggs and fully inhibits extracellular-induced NAADP Ca²⁺ release. At 1 mM, this modulator significantly reduces CCK-induced Ca²⁺ oscillations in mouse pancreatic acinar cells with minimal effect on acetylcholine-induced Ca²⁺ response. Furthermore, it completely abolishes NAADP-induced Ca²⁺ transients in intact sea urchin eggs when applied extracellularly at a concentration of 10 mM.

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
