Description
IBS008738 is an effective TAZ activator that stabilises TAZ and increases unphosphorylated TAZ levels in C2C12 cells, upregulates MyoD-dependent gene transcription, promotes muscle repair in injury models, and prevents muscle atrophy.
In vitro
IBS008738 promotes myogenesis in C2C12 cells within the concentration range of 1-10 μM, with the most significant effect observed at 10 μM. The compound exhibits optimal efficacy when administered during the initial 24 hours of differentiation, markedly enhancing the expression of myogenin and MyoD [1]. IBS008738 facilitates myogenesis and muscle repair by stabilizing TAZ and strengthening the interaction between TAZ and MyoD, thereby promoting the transcription of myogenesis-related genes [1]. Treatment with IBS008738 (10 μM, 24 hours) elevates interleukin-10 (IL-10) levels in C2C12 cells [2].
In vivo
In the mouse model of myocardial toxin-induced muscular injury, IBS008738 (3 nmol) promoted muscle regeneration and increased the number of central nucleated muscle fibers [1].
In the dexamethasone-induced muscular atrophy mouse model, IBS008738 (30 μM, 100 μL) prevented muscular atrophy and increased the cross-sectional area of muscle fibers and protein synthesis [1].
In the traumatic brain injury (TBI) mouse model, IBS008738 (30 μM, 100 μL, intramuscular injection, immediately and on day 2 after TBI) significantly reduced the expression of TNFα and IL-6 but markedly increased the expression of IL-10 [2].
