Description
CTX-471 is a fully humanized monoclonal antibody targeting CD137, exhibiting agonistic activity and binding affinity (Kd=50/61/748 nM) to recombinant human, cynomolgus monkey, and mouse CD137. It enhances T cell and natural killer cell activation, making it suitable for studying immunomodulation and cancer.
In vitro
CTX-471 (0.1-100 nM, 1 μg/mL and 10 μg/mL, 3 days) can promote IFN-γ secretion by T cells derived from Homo sapiens in an Fcγ receptor-dependent manner [1].
In vivo
Intraperitoneal administration of CTX-471 at a dose of 150 μg demonstrated curative activity as monotherapy across multiple syngeneic tumor models, exhibiting the unique advantage of achieving complete recovery in mice bearing large tumors. When administered via intravenous injection (10-80 mg/kg, on days 0, 7, 14, and 21), CTX-471 showed good tolerability with no significant signs of hepatotoxicity observed even at high doses [1].
