Description
BI-1124 is a cathepsin S inhibitor that demonstrates simulated adequate ocular pharmacokinetic-pharmacodynamic coverage when administered topically in rabbit models. It is applicable for research on age-related dry eye syndrome.
In vitro
BI-1124 (0.075 mg/mL, 0.0075% w/v) exhibits high corneal permeability and adequate solubility in computer-simulated ADMET and ocular pharmacokinetics, with an appropriate distribution coefficient at pH 7.4. Upon simulated local administration, its concentrations in rabbit cornea and conjunctiva remain above the in vitro IC 50 (7 nM) and IC 90 (70 nM) levels, supporting its potential for repurposing through cathepsin S inhibition for age-related dry eye disease.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
