Description
Anti-TNBC agent-6 (compound pt-3) is an effective agent against triple-negative breast cancer (TNBC). It exhibits cytotoxic activity and induces autophagy (autophagy) and ferroptosis (ferroptosis). Additionally, anti-TNBC agent-6 enhances intracellular ROS accumulation and demonstrates antitumor activity, offering potential for breast cancer research.
In vitro
Anti-TNBC agent-6 (compound pt-3) exhibits cytotoxic effects on MDA-MB-231, MCF-7, B16-F10, SK-OV-3, and WI-38 cells over a 48-hour period. At a concentration of 2 µM and various time points (0, 12, 24, 36, 48 h), it induces autophagy in MDA-MB-231 cells by increasing the expression of Beclin-1 and LC3-II in a time-dependent manner. Additionally, it reduces the expression of GPX4 to induce ferroptosis time-dependently. The agent also raises MDA levels in a dose-dependent manner in MDA-MB-231 cells, tested at 1 and 2 µM. Furthermore, at 2 µM, it enhances intracellular ROS accumulation and increases the labile iron pool (LIP).
In vivo
The compound anti-TNBC agent-6 (10 mg/kg; i.p.; administered on days 0, 3, 6, 9, 12, 15, and 18) exhibited antitumor activity in the MDA-MB-231 xenograft model.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
