| In vitro | Anti-TNBC agent-6 (compound pt-3) exhibits cytotoxic effects on MDA-MB-231, MCF-7, B16-F10, SK-OV-3, and WI-38 cells over a 48-hour period. At a concentration of 2 µM and various time points (0, 12, 24, 36, 48 h), it induces autophagy in MDA-MB-231 cells by increasing the expression of Beclin-1 and LC3-II in a time-dependent manner. Additionally, it reduces the expression of GPX4 to induce ferroptosis time-dependently. The agent also raises MDA levels in a dose-dependent manner in MDA-MB-231 cells, tested at 1 and 2 µM. Furthermore, at 2 µM, it enhances intracellular ROS accumulation and increases the labile iron pool (LIP). |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
