Description
ABCA1 inducer 3 (Compound 85) is an orally active ABCA1 inducer and lipid regulator. It enhances ABCA1 expression levels and upregulates mRNA expression of Abcg5 and Abcg8 in the liver. Additionally, ABCA1 inducer 3 promotes cholesterol efflux and ameliorates hyperlipidemia.
In vitro
ABCA1 inducer 3 (10 μM; 24 h) significantly upregulates ABCA1 in ABCA1p-LUC HepG2 cells by 578%, with an EC50 of 0.15 μM. In RAW264.7 macrophages, ABCA1 inducer 3 (2.5-10 μM; 24 h) moderately and dose-dependently increases Abca1 mRNA expression. Additionally, ABCA1 inducer 3 (10 μM; 18 h) enhances cholesterol efflux from RAW264.7 macrophages.
In vivo
Administering ABCA1 inducer 3 at a dosage of 50 mg per day orally for 8 weeks offers protective effects against hyperlipidemia in golden hamsters.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
