5-LOX-IN-8 is a 5-LOX inhibitor with anti-inflammatory properties. It suppresses IL-6, IL-1β, TNF-α, and IFN-γ in macrophages and reduces IL-8 secretion in SW480 cells. | In vitro | 5-LOX-IN-8 (Compound 18d) demonstrates cytotoxic effects on THP-1 cells (IC 50: 37.65 μM) and SW 480 cells (IC 50: 14.37 μM) at concentrations ranging from 0.3 to 50 μM over 24 hours. At 10 μM for 5 minutes, it inhibits human recombinant 5-LOX by 40.65% and reduces NO production in SW480 cells by 38.85%. When pre-treated at 10 μM for 1 hour followed by co-culture with LPS for 24 hours, 5-LOX-IN-8 suppresses the LPS-induced production of IL-6 (93.98%), IL-1β (19.41%), TNF-α (45.97%), and IFN-γ (66.61%) in THP-1 macrophages. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
