| Description | SMIP-031 is a potent oral PPM1A inhibitor with an IC50 value of 180 nM. It can induce autophagy (autophagy) and inhibit Mycobacterium tuberculosis infection in mice. |
| In vitro | SMIP-031, administered at concentrations of 0.5, 1, 2.5, and 5 µM over a period of 5 days, results in a dose-dependent reduction in the burden of Mycobacterium tuberculosis (Mtb) in infected THP-1 cells. Additionally, SMIP-031 at dosages of 0.25, 0.5, 1, 2, 4, and 8 µM induces autophagy by decreasing the expression of S403-p62 protein in THP-1 macrophages infected with Mtb. |
| In vivo | SMIP-031 administered intraperitoneally at doses of 12.5 and 50 mg/kg daily over a period of 8 days effectively decreased the bacterial load in the spleens of infected mice. |
