Description

RAPT-37-Me is a GCN2 inhibitor, exhibiting IC50 values for GCN2, PERK, HRl, and PKR of 0.002 μM, >10 μM, 4.40 μM, and 1.31 μM, respectively.

In vitro

RAPT-37-Me (0-10 μM, 72 h) exhibits a U-shaped activity response in MOLM-16 cells, with the most significant decrease in cell viability observed at moderate concentrations (approximately 0.01 μM), while lower and higher concentrations show reduced inhibition. EMD-C02602 (0.05-0.12 μM, 8 h) enhances ATF4 expression in MOLM-16 cells. Additionally, EMD-C02602 (0.01-0.37 μM, 8 h) increases p-GCN2 expression without affecting GCN2 expression.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSO TargetMol | reagent stock solution to 400 μL PEG300 and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH₂O TargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
