Description
PD-1/PD-L1-IN-61 is an inhibitor of the PD-1/PD-L1 interaction, with an IC50 value of 5.3 nM for human studies. It binds stably within the hydrophobic pocket of the PD-L1 dimer interface through hydrogen bonds and π-π stacking interactions with PD-L1 residues. As an immune activator, PD-1/PD-L1-IN-61 enhances the immune cytotoxicity of peripheral blood mononuclear cells against cancer cells, restores T cell immune function, and promotes IFN-γ secretion. This compound is applicable in research related to triple-negative breast cancer.
In vitro
PD-1/PD-L1-IN-61 (III-4) exhibits very low cytotoxicity towards MDA-MB-231 and Jurkat cells at concentrations of 1-16 μM. At concentrations of 0.5-2 μM, PD-1/PD-L1-IN-61 enhances PBMC-mediated cytotoxicity against MDA-MB-231 cells in a dose-dependent manner, achieving 38.4% tumor cell lysis at 1 μM. Additionally, PD-1/PD-L1-IN-61 promotes IFN-γ secretion by human peripheral blood mononuclear cells (hPBMCs) in a dose-dependent fashion at concentrations of 10-1000 nM, with significant secretion observed at 100 nM and 1000 nM.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
