Description
NAMPT inhibitor-linker 2, a drug-linker conjugate for antibody-drug conjugates (ADCs), comprises an NAMPT inhibitor payload and a linker. When combined with an anti-c-Kit monoclonal antibody to form ADC-4, it demonstrates potent efficacy against c-Kit expressing cell lines, including GIST-T1 and NCI-H526, with IC50 values of <7 pM and 40 pM, respectively.
In vitro
ADC-4 exhibits potent activity against c-Kit expressing cell lines, including GIST-T1 and NCI-H526, with IC50s of <7 pM and 40 pM, respectively[1]. ADC-4 (0.01 nM-10 nM, 144 hours) inhibits GIST-T1 cell proliferation without affecting the cell cycle[1].
In vivo
ADC-4 (20 mg/kg, i.v. for 28 days) is well-tolerated and induces tumor stasis in mice with GIST-T1 cells[1].
