JWZ-8-103 is an orally active molecular gel that targets TRIM21. It promotes the specific binding of TRIM21 to the nuclear pore protein NUP98 (EC50 ≈ 20 μM) by enhancing TRIM21’s thermal stability, and upon forming a ternary complex, it induces the ubiquitination and degradation of the nuclear pore complex. JWZ-8-103 exhibits an IC50 of 9.5 nM against pancreatic cancer cells, possesses excellent pharmacokinetic properties and good oral bioavailability, and effectively inhibits tumor growth in pancreatic cancer xenograft models.

Preparation of the In Vivo Formulation:
1) Add 100 μL of the DMSO stock solution to 400 µL PEG300 and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
