In vitro	GABAA receptor agent 8 (compound 5e) demonstrates anticonvulsant activity, evidenced by an IC50 of 0.334 μM in the 4-AP induced hyper-excitability model [1]. Additionally, this compound enhances GABA-induced activation of the GABAA1 receptor in a dose-dependent manner over a range of 0.01-100 μM and a duration of 0-140 seconds, with an EC50 of 12.02 μM.
In vivo	GABAA receptor agent 8 (compound 5e) demonstrates anticonvulsant efficacy in mice, exhibiting an effective dose (ED50) of 40.95 mg/kg and a toxic dose (TD50) of 372.39 mg/kg [1]. This agent specifically mitigates seizures induced by pentylenetetrazol (PTZ) without affecting those caused by maximal electroshock (MES) in mice [1].
