Description
FXR agonist16 is an FXR agonist with an EC50 of 2.2 μM. It activates the transcriptional activity of FXR, leading to increased expression of SHP and BSEP, while reducing the expression of Cyp7a1. FXR agonist16 demonstrates hepatoprotective activity by decreasing AST and ALT levels in hepatocyte injury models induced by free fatty acids. This compound is useful in liver injury research.
In vitro
FXR agonist 16 (Compound 3q) acts as an FXR agonist in stably transfected HEK293 cells, with an EC50 of 2.2 μM when used at concentrations of 10-100 μM for 24 hours. Additionally, FXR agonist 16 demonstrates hepatoprotective activity in FFA-damaged AML12 cells at concentrations of 1-20 μM for 24 hours, reducing liver injury markers without significant cytotoxicity up to 25 μM. Furthermore, it modulates the expression of FXR downstream target genes in AML12 cells by upregulating SHP and BSEP while downregulating Cyp7a1 at concentrations of 1-10 μM.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
