DDP-38003 trihydrochloride is a novel, orally available histone lysine-specific demethylase 1A (KDM1A/LSD1) inhibitor with an IC50 of 84 nM.

In vitro
DDP-38003 inhibits KDM1A with an IC50 of 84 nM. It is more effective in reducing colony-forming ability and inducing differentiation of THP-1 cells compared to the 1R, 2S analogue [1].
In vivo
DDP-38003 demonstrates effectiveness in vivo upon oral administration, leading to a 62% increase in survival rates in mouse leukemia models via KDM1A inhibition. It has a half-life of 8 hours. A notable dose-dependent enhancement in survival rates of mice treated with DDP-38003 is observed, with survival rates escalating by 35% and 62% at doses of 11.25 and 22.50 mg/kg, respectively, showcasing its potential as a cancer therapeutic [1].
