Bioactivity
Description
Daun02 is the topoisomerase inhibitor Daunorubicin prodrug.
In vitro
Daun02 is converted by β-galactosidase to Daunorubicin, which has been shown to reduce Ca2+-dependent action potentials in neuroblastoma cells [1]. Daun02 is a good substrate for β-galactosidase (β-gal) and it also is a topoisomerase inhibitor [2]. The concentration of Daun02 producing 50% (EC50) decrease in cell viability is 0.5 μM, 1.5 μM, and 3.5 μM for T47-D, Panc02, and MCF-7, respectively [3].
In vivo
Daun02 is an efficient substrate for β-gal (Km: 0.37 mM; Vmax: 8.6 μmol/min/mg). It demonstrates 79% plasma protein binding, compared to 94% for Daunomycin at 10 μM [3].

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
