AS-85 is a cell permeable ASH1L inhibitor with anti-leukemic activity, inhibits the growth of leukemic cells, increases cLogP, decreases tPSA, and can be used in the study of leukemia.

In vitro
AS-85 is a potent inhibitor of ASH1L histone methyltransferase with an IC50 of 0.6 µM and significant antileukemic activity.AS-85 binds tightly to the SET domain of ASH1L with a Kd value of 0.78 µM. AS-85 inhibits the proliferation of leukemia cells harboring different MLL1 translocations such as MV4;11, MOLM13 and KOPN8. AS-85 inhibited the proliferation of leukemia cells carrying different MLL1 translocations (e.g. MV4;11, MOLM13 and KOPN8) with GI50 values ranging from 5 µM to 25 µM, but did not have a significant effect on K562 leukemia cells without the MLL1 translocation. Stock Solution Preparation:

Dissolve 2 mg of the compound in 100 μL DMSOTargetMol | reagent to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
