Description | Antiparasitic agent-9 (compound 47) is a highly active and orally administered antiparasitic agent, exhibiting potent antiparasitic efficacy against the human parasite [1]. |
In vivo | Antiparasitic agent-9 (compound 47) administered at a dosage of 15 mg/kg orally, twice daily for seven days, effectively reduces the adult worm burden in the jird L. sigmodontis infection model, achieving a significant 59% reduction. When administered once at varying doses (2 mg/kg intravenously, and 10 and 30 mg/kg orally), it demonstrates a brain to plasma ratio of 0.08, indicating minimal brain penetration. Pharmacokinetic analysis in male CD-1 mice revealed that the compound, at a dosage of 30 mg/kg orally, reached a maximum concentration (C max) of 13.9 μM and had an absorption time (T max) of 0.5 hours, with an area under the curve (AUC) from 0 to infinity of 121 μM·h. Clearance (CL) was 3.6 mL/min/kg, and the steady-state volume of distribution (Vd ss) was 0.74 mL/kg, showcasing a bioavailability (F) of 57% and an efflux ratio (ER) of 12, evidencing the pharmacokinetic properties of antiparasitic agent-9. |
