Description
Antiparasitic agent-30 is an effective regulator of the adenosine kinase TbrAK and an anti-trypanosomal compound. Antiparasitic agent-30 (Compound 1) effectively inhibits the activity of the T. b. rhodesiense parasite with an IC50 of 1.0 μM. At concentrations of 1-750 μM over 10 minutes, it enhances recombinant TbrAK activity in a concentration-dependent manner, with an EC50 of 38.9 μM and showing a 2.5-fold increase in activity at 50 μM. In radiolabeled assays, its potent efficacy is demonstrated by an EC50 of 38 nM in enhancing recombinant TbrAK activity. At 150 μM, Antiparasitic agent-30 binds to two different sites on recombinant TbrAK (7 μM), showing high affinity for the primary binding site with a Kd of 75 nM. Its effectiveness decreases against TbAK-silenced T. b. brucei parasites, with the IC50 increasing from 131 nM to 271 nM, confirming that its activity is dependent on adenosine kinase expression.
