Antibiofilm agent-5 (compound 6c) is a multitarget antibacterial agent with potent antibiofilm activity. It induces metabolic dysfunction by inactivating lactate dehydrogenase, promoting the accumulation of reactive oxygen species, which subsequently reduces reduced glutathione and leads to bacterial oxidative damage. Antibiofilm agent-5 is applicable for studying recalcitrant biofilm-enhanced bacterial infections.

| In vitro | Antibiofilm agent-5 (compound 6c) exhibits superior inhibitory activity against Acinetobacter baumannii and Escherichia coli compared to norfloxacin, with a minimum inhibitory concentration (MIC) of 0.0066 mmol/L. This compound can intercalate into DNA, forming a 6c-DNA supramolecular complex that leads to cell death. Antibiofilm agent-5 shows negligible hemolytic activity on human red blood cells and exhibits low cytotoxicity to normal human liver cells (LO2) and lung epithelial cells (BEAS-2B), with no risk of crossing the blood-brain barrier.  |
