Description
AMG-34 is a GCN2-targeting agent, with IC50 values of 0.395 μM for GCN2, 0.010 μM for PERK, 1.07 μM for HRl, and greater than 10 μM for PKR.
In vitro
AMG-34 demonstrates a U-shaped activity response in MOLM-16 cells at concentrations of 0-10 μM over 72 hours, with the most significant reduction in cell viability occurring at moderate concentrations (around 0.1 μM), while both lower and higher concentrations show diminished inhibitory effects. Additionally, AMG-34 (0.01 ~ 0.37 μM, 8 h) increases the expression of ATF4 in MOLM-16 cells, and at concentrations of 1.1 ~ 10 μM over 8 hours, it reduces the expression of p-GCN2 without affecting the expression of GCN2.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
