In vitro
AM12814 (Compound 12) binds with high affinity to cannabinoid receptors, exhibiting K i values of 0.7 nM for rCB1 and 3.4 nM for hCB2. Within 30 minutes, AM12814 inhibits the Forskolin-induced accumulation of cAMP in 3xHA-hCB1-CHO and 3xHA-hCB2-CHO cells, with EC 50 values of 8.75 nM and 22 nM, respectively. After 90 minutes, the compound recruits β-arrestin 2 in hCB1-DRx-CHO and hCB2-DRx-U2OS cells, with EC 50 values of 25 nM and 5.6 nM, respectively.
In vivo
AM12814 (Compound 12), administered at a dose of 0.3-3 mg/kg via intraperitoneal injection, induces CB1-mediated cannabinoid-like behaviors in CD1 mice.
