| In vitro | Instructions for Aggregation of β-Amyloid Peptides (The following is our recommended protocol, which should be adjusted based on your specific requirements). 1. Dissolve solid Aβ peptide in cold hexafluoro-2-propanol (HFIP). Incubate the peptide at room temperature for a minimum of 1 hour to achieve monomerization and structural randomization. 2. Remove HFIP by evaporation and store the resulting peptide as a thin film at -20 or -80 °C. 3. Dissolve the resulting film in anhydrous DMSO at a concentration of 5 mM, then vortex and dilute to the desired concentration and buffer (serum-free and phenol red-free medium). 4. Age the solution at 4-8 °C for 48 hours, then centrifuge the sample at 4-8 °C and 14000g for 10 minutes; soluble oligomers will remain in the supernatant. Dilute the supernatant 10-200 fold for experimentation. Methods may vary depending on downstream applications. Note: Aggregated forms are unstable in solution and it is recommended to use them immediately. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
