Description
Werner syndrome RecQ helicase-IN-3 is a potent and orally active inhibitor of werner syndrome RecQ helicase with an IC50 value of 0.06 μM, exhibiting strong antiproliferative and anticancer activity, and serving as a targeted probe for studying WRN-dependent DNA repair mechanisms and synthetic lethal strategies in oncology research.
In vitro
In cell proliferation assays, Werner syndrome RecQ helicase-IN-3 exhibited antiproliferative activity against MSI-H colorectal cancer SW48 cells with a GI50 of 0.06 μM [1].
In vivo
In female nude mice bearing SW48 xenograft tumors (MSI-H model), oral administration of Werner syndrome RecQ helicase-IN-3 at a dosage of 120 mg/kg (once daily for 22 days) resulted in the inhibition of tumor growth. These results characterize its efficacy in targeting WRN helicase within MSI-positive cancer environments in vivo [1].
