Description
TRV056 is a Gq-biased agonist of the angiotensin II type 1 receptor (AT1R), demonstrating efficacy in stimulating Gq-mediated cellular signaling. It can serve as a foundation for the development of Gq-biased AT1R agonists.
In vitro
TRV056 is a "gain-of-function" ligand exhibiting over tenfold stronger allosteric coupling to Gq compared to Angiotensin II (AngII), thereby more effectively stimulating Gq-mediated cellular signaling, such as IP1 generation. As a Gq-biased AT1R agonist, it surpasses the endogenous ligand AngII in both efficacy of stimulating Gq-mediated signaling and in enhancing Gq coupling[1].

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
