Description
TLR8 agonist10 is a specific TLR8 agonist with an EC50 of 0.019 μM in humans. In vitro
TLR8 agonist 10 (Compound 23A) at concentrations of 0-10 μM can effectively activate latent HIV-1 in U1 cells in vitro without significant cytotoxicity. Additionally, TLR8 agonist 10 (0-10 μM) can indirectly activate latent HIV-1 in ACH-2 cells through cytokine signaling produced by treated human PBMC, also showing no significant cytotoxicity to ACH-2 cells. Furthermore, TLR8 agonist 10 (0-10 μM) potently activates latent HIV-1 in OM10.1 cells in a concentration-dependent manner with negligible cytotoxicity.
In vivo
The compound, TLR8 agonist 10 (1-5 mg/kg; IV injection, gavage; single dose), demonstrates favorable pharmacokinetic properties in male SD rats, characterized by a long half-life and low clearance. Additionally, at a gavage dose of 5 mg/kg, it achieves an oral bioavailability of 35%.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
