Bioactivity

Description	TBI-223 is an orally effective antibiotic and antibacterial agent active against Mycobacterium tuberculosis. It demonstrates efficacy against methicillin-resistant Staphylococcus aureus infections in mouse interference models and is applicable for tuberculosis research.
In vitro	In 7H9 medium, the half-maximal inhibitory concentration (MIC₅₀) values of TBI-223 against Mycobacterium tuberculosis, Mycobacterium kansasii, MAC, and MAbC were 1.50 μg/mL, 2.00 μg/mL, 8.00 μg/mL, and 2.00 μg/mL, respectively [1].
In vivo	TBI-223 (80-160 mg/kg, orally administered twice daily for 7 days) achieved a 100% survival rate in the MRSA bacteremia model of C57BL/6 mice [2]. TBI-223 (80-160 mg/kg, orally administered twice daily for 7 days) significantly reduced the infected lesion area in the MRSA skin wound infection model of both non-diabetic C57BL/6 mice and diabetic TallyHo/JngJ mice [2]. TBI-223 (80-160 mg/kg, orally administered twice daily for 7 days) significantly decreased the bioluminescence imaging (BLI) signal intensity in the MRSA orthopedic implant-associated infection model of C57BL/6 mice, while also reducing the colony-forming unit (CFU) counts on joint tissues and implant surfaces [2].

Stock Solution Preparation:

Dissolve 2 mg of the compound in 100 μL DMSO TargetMol | reagent to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSO TargetMol | reagent stock solution to 400 μL PEG300 and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH₂O TargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
