| In vitro | SB5-171 (Compound 14) effectively inhibits full-length recombinant PfCLK3, exhibiting an apparent IC50 of 10 nM under K m ATP conditions and 12 nM at 3 mM ATP. SB5-171 (100 pM-100 μM; 72 h) strongly suppresses the asexual blood stage growth of Plasmodium falciparum, with an EC50 of 104 nM for the 3D7 strain and 267 nM for the chloroquine-resistant Dd2 strain. The compound shows low cytotoxicity over 24-48 hours, with a CC50 greater than 100 μM in HepG2 cells and 25 μM in THP-1 cells. |

Preparation of the In Vivo Formulation:
1) Add 100 μL of the DMSO stock solution to 400 µL PEG300 and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
