| In vitro | N-Desmethyl xanomeline, ranging from 0.1 nM to 10 µM, exhibits intricate polypharmacological properties with strong binding potential to σ1, 5-HT 2B, 5-HT 1D, 5-HT 7, and muscarinic receptors (Ki values ranging from 6.2 nM to 219 nM). At 10 µM, it shows potential agonistic activity on various muscarinic, 5-HT, and histaminergic GPCRs. It acts as a full or partial agonist at 5-HT 1D receptors (potency range 17-130 nM) and 5-HT 1A receptors (EC 50 = 2.20 µM), while at 5-HT 2A, 5-HT 2C, H 3, and H 4 receptors, it is a weak partial agonist. |

Preparation of the In Vivo Formulation:
1) Add 100 μL of the DMSO stock solution to 400 µL PEG300 and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
