Lithium orotate is an orally effective lithium supplement with low binding affinity, allowing it to bypass amyloid chelation in Alzheimer's disease (AD) mouse models. In these models, as well as in aged wild-type mice, lithium orotate prevents the deposition of Aβ plaques and the accumulation of phosphorylated tau proteins. Description
Lithium orotate is an orally effective lithium supplement with low binding affinity, allowing it to bypass amyloid chelation in Alzheimer's disease (AD) mouse models. In these models, as well as in aged wild-type mice, lithium orotate prevents the deposition of Aβ plaques and the accumulation of phosphorylated tau proteins. It also reverses AD pathology, neuroinflammatory changes, and memory loss. Furthermore, lithium orotate is applicable in the research of alcoholism and Alzheimer's disease.
In vitro
Lithium orotate (0-500; 16h) shows limited binding affinity to synthetic human Aβ 1-42 fibrils and oligomers. Additionally, lithium orotate (20-500 μM; pretreatment
6h) enhances the uptake and degradation of Aβ 42 by BV2 microglial cells.
In vivo
Lithium orotate (4.3-430 µM; oral administration; drinking water intake) effectively prevents and reverses pathological changes of Alzheimer's disease and cognitive decline in AD mouse models as well as in aging wild-type mice.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
