Lipid I97 is an ionizable cationic lipid derived from vitamin B5 with a pKa of 6.23. It has been utilized in the formation of lipid nanoparticles (LNPs) for mRNA delivery in vivo. In mice, LNPs with lipid I97 encapsulating luciferase mRNA specifically accumulate in the spleen and lymph nodes. Additionally, LNPs containing lipid I97 with mRNA encoding hemagglutinin A trigger the production of IgG1 and IgG2 in mice.
