Influenza HA (518-526) is an H-2d-restricted CD8+ T cell epitope derived from the influenza virus hemagglutinin. Description
Influenza HA (518-526) is an H-2d-restricted CD8+ T cell epitope derived from the influenza virus hemagglutinin. This sequence is highly conserved across multiple H5N1, some H9N2, and certain H1N1 strains. The epitope can bind to the mouse MHC class I allele Kd to form a complex, which is subsequently recognized by specific CD8+ T lymphocytes. In influenza-infected BALB/c mice, Influenza HA (518-526) represents an immunodominant epitope that stimulates CD8+ T cells to release IFN-γ, thereby inducing a robust immune response. Currently, this epitope has been widely applied in research related to respiratory syncytial virus (RSV), influenza virus, and H5N1 influenza, among other fields.
In vitro
Method: Influenza HA (518-526) was inserted into the stalk region of neuraminidase (NA) of PR8 influenza virus to construct a recombinant virus. NA enzyme activity was measured after virus purification. MDCK cells were infected at an MOI of 0.001, virus growth kinetics were assessed at 48 h post-infection, and plaque size was observed at 72 h post-infection.
Result: After insertion of Influenza HA (518-526), the NA enzyme activity of the virus decreased to 15% of that of the wild-type virus . However, the virus growth kinetics (at 48 h) and plaque size (at 72 h) in MDCK cells showed no significant difference from those of the wild-type virus [1].
In vivo
Method: BALB/c mice were intranasally inoculated with PR8/NA-HA₅₁₈₋₅₂₆ virus, and body weight changes were recorded daily to evaluate pathogenicity. Spleen cells were isolated 10-14 days post-infection and restimulated with Influenza HA (518-526) peptide, and CTL responses were detected by IFN-γ ELISPOT and flow cytometry.
Result: After integration of Influenza HA (518-526) into the neuraminidase stalk of PR8 influenza virus, the pathogenicity of this recombinant virus in mice was weaker than that of the wild-type PR8 virus. It induced Influenza HA (518-526)-specific CTL responses but did not induce RSV F₈₅₋₉₃-specific CTL responses [1].
