Description
HSV-gB2 (498-505) is an immunodominant epitope derived from glycoprotein B of herpes simplex virus (HSV), corresponding to amino acid residues 498 to 505 of the protein. This epitope serves as a recognition target for cytotoxic T lymphocytes (CTLs) that are H-2Kb-restricted and cross-reactive against HSV-1 and HSV-2.
In vivo
Method: C57BL/6 mice were immunized once in the rear footpads with 200 μg of the gB498-505 synthetic peptide (emulsified in TiterMax adjuvant). Eight days later, popliteal lymph node cells were harvested and cultured in vitro for 3 days without antigenic stimulation. Cytolytic activity was assessed using a ⁵¹Cr release assay against B6/WT-3 target cells either infected with HSV-1 or pulsed with the gB498-505 peptide. The phenotype of the effector cells was identified by treatment with antibody and complement.
Results: Lymph node cells from immunized mice acquired the ability to specifically lyse both HSV-1-infected and gB498-505-pulsed target cells in the absence of in vitro antigenic restimulation. This lytic activity was dependent on CD8⁺ T cells, whereas depletion of CD4⁺ T cells did not reduce the killing effect. These findings indicate that gB498-505 can induce the differentiation of CD8⁺ CTLs in vivo to an antigen-independent maturation stage [1].
