| In vivo | Method: Allergic contact dermatitis was induced in BALB/c mice by topical application of dinitrochlorobenzene (DNCB) or toluene-2,4-diisocyanate (TDI). Diflorasone diacetate (0.05%, 10 μg/20 μL) was applied locally to the right ear 2 hours before challenge. Ear thickness, inflammatory cell infiltration, and chemokine (RANTES, TARC, CTACK) concentrations in ear tissue homogenates were measured 24 hours after challenge using ELISA. Infiltration of CD4⁺ T cells, CD8⁺ T cells, and neutrophils was assessed by immunohistochemistry. Result: Topical pretreatment with Diflorasone diacetate significantly inhibited TDI- and DNCB-induced ear swelling (P < 0.05) and markedly reduced the infiltration of CD4⁺, CD8⁺ T cells, and GR-1⁺ neutrophils at the inflamed site [16]. It significantly reduced elevated RANTES levels in both models. In the DNCB model, it suppressed the increase in TARC, whereas in the TDI model, it paradoxically led to a significant increase in TARC concentration (P < 0.05). No significant effect on CTACK levels was observed [1]. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 µL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
