Description
Diflorasone diacetate is a topical anti-inflammatory steroid preparation used in dermatological research to alleviate corticosteroid-responsive skin diseases.
In vivo
Method: Allergic contact dermatitis was induced in BALB/c mice by topical application of dinitrochlorobenzene (DNCB) or toluene-2,4-diisocyanate (TDI). Diflorasone diacetate (0.05%, 10 μg/20 μL) was applied locally to the right ear 2 hours before challenge. Ear thickness, inflammatory cell infiltration, and chemokine (RANTES, TARC, CTACK) concentrations in ear tissue homogenates were measured 24 hours after challenge using ELISA. Infiltration of CD4⁺ T cells, CD8⁺ T cells, and neutrophils was assessed by immunohistochemistry.
Result: Topical pretreatment with Diflorasone diacetate significantly inhibited TDI- and DNCB-induced ear swelling (P < 0.05) and markedly reduced the infiltration of CD4⁺, CD8⁺ T cells, and GR-1⁺ neutrophils at the inflamed site [16]. It significantly reduced elevated RANTES levels in both models. In the DNCB model, it suppressed the increase in TARC, whereas in the TDI model, it paradoxically led to a significant increase in TARC concentration (P < 0.05). No significant effect on CTACK levels was observed [1].

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
