Description
COX-2/NO-IN-1 is an orally active nitric oxide synthase (iNOS) and NO (IC50 of 3.52 μM) inhibitor that also suppresses COX-2 expression, exhibiting anti-inflammatory effects and reducing pathological damage to kidney tissues [1].
In vitro
COX-2/NO-IN-1 (Compound 13b; 2.5-10 μM) exhibits notable anti-inflammatory effects by inhibiting nitric oxide (NO, IC50 = 3.52 μM), TNF-α, and IL-6 production (84.1% and 33.6%, respectively) in a dose-dependent manner, and reducing the expression of iNOS, COX-2, and TLR4 proteins in LPS-stimulated RAW264.7 cells [1].
In vivo
In C57BL/6 mice with cisplatin-induced AKI, COX-2/NO-IN-1 (Compound 13b; 15–30 mg/kg; p.o.) effectively maintains normal metabolic function, inhibits inflammation, and mitigates kidney tissue pathological damage.
