In vitro
Antiparasitic agent-28 (Compound 12) is a potent inhibitor of Toxoplasma gondii tachyzoite growth (TgME49-Fluc strain) with an EC50 of 1 nM and an EC90 of 2 nM after 72 hours. At 96 hours, it effectively inhibits alkaline-induced T. gondii bradyzoites (Tg68nLuc strain) with an EC50 of 3 nM and glutamine-rich medium-induced bradyzoites with an EC50 of 0.1 nM. This compound exhibits low toxicity toward human liver HepG2 cells (CC50 = 0.99 μM, SI = CC50/EC50 = 1447) and human monocyte THP-1 cells (CC50 = 0.25 μM, SI = 374).
In vivo
Antiparasitic agent-28 (Compound 12) administered orally at a dose of 3 mg/kg twice daily for 7 days can prevent chronic infection caused by TgME49-Fluc tachyzoites in female C57/BL6 mice. Additionally, Antiparasitic agent-28 at the same dosage given for 14 days significantly extends the survival of interferon γ receptor 1 knockout mice infected with TgME49-Fluc tachyzoites.
