Description
Antiparasitic agent-25 (Compounds 11) is an orally active antiparasitic compound. It inhibits the invasion and replication capabilities of parasites and has an irreversible effect on Toxoplasma gondii. The agent significantly reduces the replication rate of Toxoplasma gondii, with an IC50 value of 6.33 μM, and exhibits low cytotoxicity, having a CC50 value of 285 μM.
In vitro
Antiparasitic agent-25, when applied at a concentration of 10 μM, exhibits irreversible effects on Toxoplasma over a period of 24 to 72 hours. Furthermore, a 30-minute exposure to this compound significantly reduces the number of Toxoplasma invading host cells, suggesting an inhibition of parasite invasion. In a 24-hour cell proliferation assay using HFF cells with the same 10 μM concentration, the majority of tachyzoites are restricted to 0-1 replication cycles, with vacuoles predominantly accommodating only one or two tachyzoites.
In vivo
Antiparasitic agent-25 (50 mg/kg; orally administered twice daily for 5 consecutive days) significantly reduces parasite load and statistically extends the median survival time. However, it is insufficient to completely inhibit parasite growth within the body, resulting in persistent infection and consequently higher mortality rates in infected mice.
