Description
Angiotensin II 5-valine acts as an agonist at the angiotensin receptor.
In vivo
Method: Male Sprague-Dawley rats underwent uninephrectomy and were continuously infused with Angiotensin II 5-valine (40 ng/min) via subcutaneously implanted osmotic minipumps for 12 days. Some rats were concurrently treated with the AT1 receptor antagonist losartan (30 mg/kg/d) in drinking water. Exogenous Angiotensin II 5-valine was separated from endogenous Angiotensin II 5-isoleucine by high-performance liquid chromatography, and peptide contents in plasma and kidney were measured by radioimmunoassay.
Result: Infusion of Angiotensin II 5-valine for 12 days significantly elevated systolic blood pressure (to 197±7 mmHg), an effect completely prevented by losartan. Renal Angiotensin II 5-valine content was markedly increased (256±44 fmol/g), with a kidney-to-plasma ratio of 6.78±1.73, indicating active renal uptake of circulating Angiotensin II 5-valine. Losartan reduced renal Angiotensin II 5-valine content by 88%, demonstrating that this uptake process is primarily mediated by AT1 receptors [1].
