AF151 effectively degrades METTL3 protein in a time-dependent manner and significantly inhibits the growth of MOLM-13 cells (DC₅₀ = 0.43 μM, IC₅₀ = 0.45 μM) at concentrations of 0-10 μM over 0-72 hours. Furthermore, AF151 at 2 μM reduces m6A levels in MOLM-13 cells within 24-48 hours. At 0.1-5 μM over 48 hours, AF151 induces apoptosis in a dose-dependent manner and decreases anti-apoptotic protein levels in these cells. Over 72 hours, AF151 and Venetoclax demonstrate synergistic effects in MOLM-13 cells. Additionally, at 0.001-10 μM concentrations, AF151 enhances the interaction between METTL3 and VHL, with binding affinities of 363 nM for AF151 and 79.9 nM for AF151-METTL3.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSO TargetMol | reagent stock solution to 400 μL PEG300 and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH₂O TargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
