AF151 is a METTL3 PROTAC degrader with a DC50 of 0.43 μM in MOLM-13 cells. It inhibits cell growth by significantly degrading METTL3 protein and reducing m6A levels. | In vitro | AF151 effectively degrades METTL3 protein in a time-dependent manner and significantly inhibits the growth of MOLM-13 cells (DC₅₀ = 0.43 μM, IC₅₀ = 0.45 μM) at concentrations of 0-10 μM over 0-72 hours. Furthermore, AF151 at 2 μM reduces m6A levels in MOLM-13 cells within 24-48 hours. At 0.1-5 μM over 48 hours, AF151 induces apoptosis in a dose-dependent manner and decreases anti-apoptotic protein levels in these cells. Over 72 hours, AF151 and Venetoclax demonstrate synergistic effects in MOLM-13 cells. Additionally, at 0.001-10 μM concentrations, AF151 enhances the interaction between METTL3 and VHL, with binding affinities of 363 nM for AF151 and 79.9 nM for AF151-METTL3. |
